種間雑種における不和合の分子基盤

Molecular basis of interspecific hybrid incompatibility

異なる種のゲノムは、なぜ一つの種子の中で共存できないことがあるのか。親由来アレルを区別したオミクス解析から、胚乳で生じる種間障壁の分子実体を明らかにします。

Why can genomes from different species sometimes fail to coexist within a single seed? Allele-resolved multi-omics enables us to define the molecular basis of interspecific barriers arising in the endosperm.

近縁な植物種を交雑すると、受精そのものは成立しても、胚乳発生が正常に進まず、種子が致死となる場合があります。胚乳は母由来2コピーと父由来1コピーのゲノムをもつため、親ゲノム間の量的バランスやエピゲノム状態のずれに特に敏感です。私たちは、栽培イネと野生イネの交雑系を用い、雑種胚乳で生じる発生異常を引き起こす分子機構の解明を目指しています。

私たちは親由来アレルごとの遺伝子発現とエピゲノム制御に着目し、インプリント遺伝子やトランスポゾン、低分子RNAなどとの関係を研究しています。さらに、胚乳の組織学的表現型と時系列オミクスを対応づけることで、原因となる初期のエピゲノム変化と、発生停止に伴って二次的に生じる変化を区別します。

これらの解析から、親ゲノム間のエピゲノム非対称性が、どの制御経路を介して胚乳発生を変化させるのかを明らかにします。また、倍数性操作や特定の遺伝的背景によって種子生存性が回復する条件を比較することで、種間障壁を形成する因子と、それを回避できる分子条件を同定します。野生イネがもつ有用形質を育種へ導入するための基礎原理の確立を目指しています。

Crosses between closely related plant species can achieve fertilization yet produce lethal seeds because endosperm development fails to proceed normally. The endosperm carries two maternal genome copies and one paternal copy, making it particularly sensitive to dosage balance and mismatched epigenomic states between the parental genomes. Using crosses between cultivated and wild rice, we aim to uncover the molecular mechanisms that cause developmental abnormalities in hybrid endosperm.

We focus on parent-of-origin-specific gene expression and epigenomic regulation, examining their relationships with imprinted genes, transposable elements, and small RNAs. By integrating histological phenotypes of the endosperm with time-resolved omics data, we distinguish early epigenomic changes that contribute to incompatibility from secondary changes associated with developmental arrest.

Through these analyses, we seek to determine how epigenomic asymmetry between parental genomes alters endosperm development through specific regulatory pathways. We also compare conditions in which seed viability is restored by ploidy manipulation or particular genetic backgrounds to identify both the factors that establish interspecific barriers and the molecular conditions that bypass them. Our goal is to establish a mechanistic foundation for introducing useful traits from wild rice into breeding programs.
